Public Abstract:
Research Objective

Generating human liver using pig as a bioreactor

Impact

The livers produced will be used for transplantation into patients with end-stage liver disease, metabolic disorders, and for metastatic liver disease not amenable to resection

Major Proposed Activities

  • MILESTONE 1 TASKS
    1) Procure liver from control pig
    2) Place on XVIVO Perfusion Device
  • MILESTONE 2 TASKS
    1) Procure livers from 5 control pigs and 5 modified pigs
    2) Perfuse on XVIVO device for up to 5 days each and collect blood and tissue samples
  • MILESTONE 3 TASKS
    1) Procure livers from 5 additional control pigs and 5 modified pigs
    2) Interim analysis of gross and histologic findings to determine the presence of accelerated and acute rejection
  • MILESTONE 4 TASKS
    1) Procure livers from up to 10 additional control pigs and 10 modified pigs as indicated by preliminary analysis
    2) Complete all blood and tissue collection
  • MILESTONE 5 TASKS
    1) Analysis of all data and statistical comparison of labs, the viability and serologic results
    2) Determine if any additional tests are required utilizing the repository samples
  • MILESTONE 6 TASKS
    1) Completion of all reports, manuscripts and presentations
    2) Preparation of protocols for future studies indicated by results of this study
Statement of Benefit to California:
There is a nationwide shortage of suitable transplant organs and California is one of the most heavily impacted states. California has one of the most profound organ shortages resulting in sicker patients being transplanted with longer hospital stays, more morbidity and impacting mortality. Significantly, Hispanics and minorities are the most disaffected grouping due to late diagnosis and referrals, often resulting in poor prognosis and moving them into high risk bracket for transplantation.
